Around-the-objective total internal reflection fluorescence for the upright (top) and inverted (bottom) microscope. A conditioned laser beam propagates through the submillimeter gap created by the oil immersion objective and glass coverslip. The slightly red pencil of light inside the prisms is the laser exciting autofluorescence. For details see Burghardt et al., pp. 6120-6131.
